Corynebacterium glutamicum KCCM 80388 utilizes fermentation to generate L-isoleucine
The European Union Reference Laboratory (EURL) has recommended a chromatographic method for the accurate determination of L-isoleucine in feed additives, premixtures, compound feed, and water. This method, which involves ion-exchange chromatography with post-column derivatization and photometric detection, is the standard procedure for amino acid quantification in feed-related samples, including water used for feed supplementation.
The recommended method aligns with regulatory requirements for precision, reproducibility, and reliability in determining free amino acids such as L-isoleucine in complex feed matrices. The EURL's endorsement ensures compliance with EU feed additive regulations.
The feed additive in question contains two products with minimum L-isoleucine content of 90.0% and 65.0% (w/w, based on dry matter). The feed additive, intended for use in feed through premixtures or directly into compound feed and water, is under authorization for L-isoleucine produced by fermentation with Corynebacterium glutamicum KCCM 80388.
For the identification of L-isoleucine in the feed additive, the EURL recommends the Food Chemical Codex (FCC) "L-isoleucine monograph" for a minimum of 90% purity, while the FCC monograph is valid for the identification of L-isoleucine in the feed additive with a minimum of 65% purity.
The determination of isoleucine in the feed additive, premixtures, and compound feed is proposed to be done using the ring-trial validated EU method based on ion-exchange chromatography (IEC) coupled to post-column derivatisation and optical (VIS) detection. On the other hand, the EU method based on IEC-VIS is recommended for the determination of isoleucine in water.
Further testing or validation of the methods is not considered necessary, as they have been extensively validated for various amino acids, including isoleucine. The EURL considers the method EN ISO 17180 valid for the determination of isoleucine in water in the current application, and the EU method (or equivalent EN ISO 13903 method) is recommended for the determination of isoleucine in feed additives, premixtures, compound feed (for compound feed only when used as a nutritional additive), and water.
Official control of the feed additive involves the FCC "L-isoleucine monograph" for the identification of L-isoleucine, the EN ISO 17180 method based on IEC-VIS/FLD for the determination of isoleucine, and the EU method based on IEC-VIS for the determination of isoleucine in water.
